I give him benadryl at night and since yesterday polysporin drops but no change.... No pu nor symptoms but now I find it worrying... pharmacy or optometrist?

I would start by stopping the Polysporin, and indeed I would advise you to consult an optometrist to get the right diagnosis and not to delay the right treatment, if any.
